چکیده
Fetal therapies regularly result in iatrogenic preterm premature rupture of the fetal membranes (iPPROM), which is associated with birth. Biomaterials that promote healing traumatized (FMs) may prevent iPPROM-associated births, addressing this unmet clinical need. Here, a fully defined synthetic poly(ethylene glycol) (PEG) hydrogel developed to study functions human amnion-derived mesenchymal stromal cells (hAMCs) 3D cultures. A pipeline analyze extracellular matrix (ECM) proteins deposited by hAMCs PEG hydrogels established involving label-free quantification mass-spectrometry data. Owing contaminant-free and short bovine serum (FBS)-free culture period, 128 ECM proteins, 97 are present native amnion, identified. Upon stimulation platelet-derived growth factor BB (PDGF-BB), cell proliferation migration inducing factor, remodel their surroundings deposit pericellularly. Among most abundantly amnion transforming β-induced protein ig-h3 (TGFβi), very distinctive involved wound cascade, These data support potential PDGF-BB repair FMs encourage its use for engineering biomaterials FM healing, ultimately iPPROM.

Identification of Extracellular Matrix Components and Biological Factors in Micronized Dehydrated Human Amnion/Chorion Membrane
Objective: The use of bioactive extracellular matrix (ECM) grafts such as amniotic membranes is an attractive treatment option for enhancing wound repair. In this study, the concentrations, activity, and distribution of matrix components, growth factors, proteases, and inhibitors were evaluated in PURION® Processed, micronized, dehydrated human amnion/chorion membrane (dHACM; MiMedx Group, Inc....
متن کاملAntibacterial Effect of Human Amnion Membrane
Abstract Background and Objective: Along with antibiotics, the use of biological methods to combat bacteria is notably considered. A natural barrier such as amniotic membrane is one of the ways of dealing with bacterial infections. The aim of this study was to determine the antibacterial effect of human amniotic membrane. Materials and Methods: This descriptive study was performed in Dezyani te...
متن کاملMatrix interactions 23 Deposition of stromal and basal laminal matrix by human amnion epithelial cells
In placental membranes near gestational term, amnion epithelial cells interact with a highly convoluted basal lamina beneath which is an acellular 'compact layer' of collagenous stroma rich in type III collagen. We have examined this extracellular matrix by SEM after fractioning the tissue or removing the epithelial cells enzymically, by TEM and by immunofluorescence with antibodies to matrix m...
متن کاملMicromorphologic changes in human amnion cells during trypsinization.
Trypsinization is a well established procedure in tissue culture technics; the trypsin effects a separation of the individual living cells from the tissue, thus permitting the preparation of suspensions of cells suitable for continued growth in culture. The effect upon the cells generally has been considered a surface phenomenon (7), although recently Levan and Biesele (6) suggested that trypsi...

Cells. Primary cultures of human amnion cells were prepared by trypsinizing amniotic membranes. Cultures were grown in Medium 5 12 (5) supplemented with 15% fetal bovine serum, penicillin (100 units/mi), and streptomycin (100 @ig/ml). Following SV4O infection, the cultures were maintained in McCoy's Medium SA (15) containing 10% agamma newborn calf serum. Approximately 6 weeks after infection, ...
